{"id":3574,"date":"2021-11-05T00:59:33","date_gmt":"2021-11-05T00:59:33","guid":{"rendered":"https:\/\/dinpertanpangan.demakkab.go.id\/?p=3574"},"modified":"2021-11-05T00:59:34","modified_gmt":"2021-11-05T00:59:34","slug":"perbanyakan-agens-hayati-golongan-cendawan-dan-bakteri","status":"publish","type":"post","link":"https:\/\/dinpertanpangan.demakkab.go.id\/?p=3574","title":{"rendered":"Perbanyakan Agens Hayati Golongan Cendawan dan Bakteri"},"content":{"rendered":"\n<p><strong>Golongan Cendawan<\/strong><\/p>\n\n\n\n<p><strong>Bahan dan Alat :<\/strong><\/p>\n\n\n\n<p>Beras\/jagung, Air bersih, Dandang, Kantong plastik tahan panas ukuran 1 kg, Stater cendawan AH, Sendok teh, Spirtus, Alkhol 70%, Hand case, Lemari pendingin dan Lampu bunsen.<\/p>\n\n\n\n<p><strong>Cara kerja :<\/strong><\/p>\n\n\n\n<ul class=\"wp-block-list\"><li>Beras\/jagung dicuci dengan air bersih selanjutnya direndam selama 15 menit, setelah itu ditiriskan sampai kering (kering angin).<\/li><li>Beras\/jagung dikukus di dandang sampai setengah matang selama 20-30 menit.<\/li><li>Beras\/jagung setengah matang diangkat didinginkan selama 5 menit. atau beras di rendam 24 jam lalu di cuci, kering anginkan.<\/li><li>Masukkan beras\/jagung dalam kantong plastik tahan panas sebanyak 100 gr lalu dilipat-lipat<\/li><li>Media beras\/jagung tersebut dikukus menggunakan dandang selama 2 jam dengan autoclave pada tekanan 1 atm, suhu 120 derajat selama 15-20 menit<\/li><li>Media beras\/jagung yang sudah steril tersebut dikeluarkan dari autoclave\/dandang lalu didinginkan<\/li><li>Setelah dingin inokulasikan (starter) dengan menggunakan sendok teh kecil steril\/jarum oxe yang dilakukan dalam lemari isolasi (hand case)<\/li><li>Inkubasikan dalam ruang bersih dengan suhu kamar.<\/li><li>Media padat yang ditutupi meselia berwarna putih (Beauveria bassiana) dan warna hijau (Metarhizium sp) yang berumur antara 2-3 minggu, siap digunakan untuk aplikasi.<\/li><\/ul>\n\n\n\n<p>Media padat yang belum digunakan langsung dapat disimpan dalam lemari&nbsp; pendingin.<\/p>\n\n\n\n<p><strong>Golongan Bakteri<\/strong><\/p>\n\n\n\n<p><strong>Bahan dan Alat<\/strong><strong>&nbsp;:<\/strong><\/p>\n\n\n\n<p>Starter Bakteri, Panci, Gelas Ukur, Pisau, Kompor, Fermentor Sederhana, Kentang 300 Gr, Gula 15 Gr, Air 1 Liter.<\/p>\n\n\n\n<p><strong>Cara kerja :<\/strong><\/p>\n\n\n\n<ul class=\"wp-block-list\"><li>Pilih kentang yang bermutu baik dan sehat.<\/li><li>Kupas kentang, kemudian cuci bersih dan diiris tipis-tipis.<\/li><li>Irisan kentang masukkan kedalam panci yang telah diberi air.<\/li><li>Rebus irisan kentang sampai empuk (jika dirasakan hambar).<\/li><li>Saring rebusan kentang dan ambil ekstraknya, masukkan ekstrak kentang ke dalam panci, tambahkan gula pasir kemudian masak dengan api kecil sampai gula larut.<\/li><li>Dinginkan ekstrak kentang gula tersebut di wadah perbanyakan<\/li><li>Masukkan stater\/isolat\/biang Bakteri<\/li><li>Nyalakan fermontor sederhana tersebut yang telah dirangkai.<\/li><li>Inkubasikan selama 5-7 hari.<\/li><\/ul>\n\n\n\n<p><strong>Sumber:<\/strong> <a href=\"http:\/\/cybex.pertanian.go.id\/artikel\/99266\/perbanyakan-agens-hayati-golongan-cendawan-dan-bakteri\/\">http:\/\/cybex.pertanian.go.id\/artikel\/99266\/perbanyakan-agens-hayati-golongan-cendawan-dan-bakteri\/<\/a><\/p>\n","protected":false},"excerpt":{"rendered":"<p>Golongan Cendawan Bahan dan Alat : Beras\/jagung, Air bersih, Dandang, Kantong plastik tahan panas ukuran 1 kg, Stater cendawan AH, Sendok teh, Spirtus, Alkhol 70%, Hand case, Lemari pendingin dan Lampu bunsen.
